Specific-immuno-(chymo) trypsin-inhibitor-assays for determination of (residual) activity of Bowman-Birk or Kunitz soyabean trypsin inhibitors

    Abstract

    Proteinaceous protease inhibitors are the major antinutritional factors in soyabeans. Accurate measurement of the protease inhibitor activity is very important in soyabean processing to yield a high-quality product for human and animal nutrition. A test principle that meets these characteristics is described. The principle is based on the binding between enzyme and functional inhibitor followed by the sensitive and accurate determination of the bound inhibitor by specific antibodies. Methods based on this principle can be performed in microtitre plates. Data obtained compare well with those from conventional trypsin inhibitor assays. The most interesting feature is the possibility of assessing the specific characteristics and properties of the different inhibitors, Kunitz soyabean trypsin inhibitor and Bowman-Birk inhibitor, in response to a variety of conditions, such as heat treatment and extrusion cooking. In this way, the inactivation kinetics of individual trypsin inhibitors (classes or iso-inhibitors) can be easily be determined.
