Induction of a tyrosinase mRNA in Agaricus bisporus upon treatment with a tolaasin preparation from Pseudomonas tolaasii

    Abstract

    In Agaricus bisporus infected with Pseudomonas tolaasii, or treated with a partially purified tolaasin extract or with purified tolaasin, induction of a specific mRNA of the two sequences encoding tyrosinase was found. Tyrosinase isoforms in samples treated with several concentrations of a partially purified tolaasin (PT) extract were visualized after Western analysis and staining with polyclonal tyrosinase-specific antibodies. Similar concentration of a 67 kDa latent tyrosinase was shown in all the samples including the water-treated control but the treatment with the PT extract caused the post-translational modification of the protein into an active tyrosinase of 43 kDa.
