Glucomannan Plus Spirulina-Enriched Squid Surimi Prevents Ampk Inhibition On Zucker FA/FA Rats Fed Hyperlipemic Diets

Abstract

Introduction AMP-activated protein kinase (AMPK) is an important metabolic enzyme, closely linked to metabolic syndrome (MS). Reactive oxygen species originating during development of diabetes inhibit AMPK phosphorylation and thus its protective effect against ß-cell dysfunction. The goal of the present study was to assess the capacity of a glucomannan-enriched/glucomannan plus spirulina-enriched surimi into a high-fat diet to increase AMPK phosphorylation in a common model for MS. Patients (or Materials) and Methods The present study was approved by the Spanish Science and Technology Advisory Committee. Three groups of 8 rats were fed for 7 weeks with control-surimi (C), glucomannan-surimi (G), or glucomannan plus spirulina-surimi (GS) diets (70% high-caloric AIM-93N modified diet (11.5% palm olein), adding 30% of freeze-dried restructured squid surimi (with 15% of glucomannan in G + 3 g/kg of spirulina powder in GS). After killing the rats, glucose, insulin, glutathione redox index, and AMPK phosphorylation levels were measured. Results According to glucemia, all rats became diabetic, but both fiber-enriched diets lowered glucose levels and improved glutathione status. The G diet decreased AMPK phosphorylation, but the GS diet induced higher insulin levels and pAMPK/AMPK values with respect to the control. Those results, together with previous ones regarding antioxidant effects of these diets, suggest the potential functional activity of GS on diabetes and MS. Concluson Glucomannan into a surimi matrix did not improve AMPK phosphorylation, but the addition of 3 g/kg of spirulina increased insulin levels and improved AMPK status in Fa/Fa rats fed a diabetogenic diet. Further studies are needed to confirm the antidiabetogenic results of this functional food.
