Description

Egg samples were collected and identified to species. Visual identification was checked with genetic identification. This project showed that it is possible to identify fish eggs to species in a mixed sample, containing multiple eggs and species.

Ethanol was removed from the eggs. Subsequently, DNA was extracted as described by Jérôme et al., 2003b. The method is based on magnetic bead technology (ChargesSwitch® Forensic DNA Purification Kit, Invitrogen, Carlsbad, USA).  A fragment of the nuclear rhodopsin gene was amplified using rhodopsin primers, described by Sevilla et al. 2007. Sequences from teleost fish species are available in the NCBI data base. Sequences were analyzed using the Qiime software package.

Currently genetic and visual identifications are compared.
