16 rats were mated and the dams continued pregnancy (controls) or were subsequently caloric restricted (CR) for 20% during days 1-12. Control female/male offspring continued normal lactation, while offspring of CR-treated dams received either normal lactation (CR group) or received during lactation until PN21 leptin supplementation. Leptin treatment of offspring during lactation after caloric restriction of dams during pregnancy reverts CR-induced dysfunction. Overall design: 16 rats were mated and the dams continued pregnancy (controls) or were subsequently caloric restricted (CR) for 20% during days 1-12. Control female/male offspring continued normal lactation, while offspring of CR-treated dams received either normal lactation (CR group) or received during lactation until PN21 leptin supplementation (physiological doses given on subsequent days: 1.0; 2.0; 3.0; 4.0; 5.0; 6.3; 7.5; 8.8; 10.0; 11.3; 15.6; 17.2; 18.8; 20.3; 21.9; 23.5; 25.0; 26.6; 39.4; 43.8 ng of leptin; CR-L group. These doses represent five times the average amount of daily leptin intake from the mother's milk). After four months ad libitum feeding with chow, a blood sample was drawn and PBMC were isolated. Snap frozen in liquid nitrogen and used for RNA isolation and whole genome gene expression microarray hybridisation using Agilent arrays.
