Description
We recorded streak camera images of WT and mutant Arabidopsis thaliana leaves to resolve the fluorescence contributions of PSI and PSII to the total fluorescence. The mutants are lhcb2 RNAi and lhca knockout. For correction and interpretation of the data, we recorded the IRF of the streak camera set-up and the decay spectra of isolated LHCII and freshly isolated grana. Next to that, we performed pulse-amplitude measurements to determine the quantum yield of PSI and PSII at increasing light intensities. We used the same technique to monitor the ability for state transitions of the Arabidopsis leaves.
